The dither looks good to me. I think the lines should not be black. Maybe make the lines dark green or dark brown.
Mushrooms have "gills" under their caps.

Also, try making the mushrooms have a shadow under their cap.

If you want to learn the art form and improve your pixel technique, it would help greatly to reduce the size of the piece. If you’ve just started out painting, you wouldn’t begin with a 4x7 meter canvas, right? I’d shrink it down to no more than 50% of its current size. There’s not enough contrast in your palette, increase the value jumps between your colors. The curves of the mushrooms look like the tops of hexagons, smooth out the points at the top and the shoulders on the sides of the large mushroom’s cap. Don’t worry about dithering yet. The most common rookie mistake is to overuse it. If necessary, do some dithering/texturing towards the end of the piece, your main concern at this point should be general construction and not the finer details. Light source seems confused.

Are you using any references? http://americanmushrooms.com/gallery.htm - This site has heaps of mushroom images. Also, ../pixelart/33605.htm - this is a good example of a PJ gallery mushroom.
I wouldn't use any dithering here; it is unnecessary (at least for the time being). Focus on defining a lightsource and try to think about the form of the objects.
Your mushrooms (and ground) are very pointed/jaggy at the moment, I would try to smooth out the line art to make things rounder/smoother. Better yet, start by blocking out general shapes instead of using lines to define everything.
The colours could also use some work, both in terms of contrast and hue-shifting. So, for example, don't just use red for the top of the mushroom, consider using oranges/purples too.

If I were you I'd blend the bottom of the stipe with the ground a bit more, it looks somewhat disconnected to me.

Well, the idea of hue shifting is to use multiple hues together, rather than creating several separate image each with different hues.
Also, the colours are hardly the greatest concern here, why not work on the rest of the piece's issues?

I still think the dithering is mostly unnecessary here. If you haven't seen it already, Cure talks a bit about dithering in this http://www.pixeljoint.com/forum/forum_posts.asp?TID=11299&PID=139322 - thread .
Also, you haven't really addressed the issues with your light source. Have a look at the lighting in http://americanmushrooms.com/taxa/Tylopilus_williamsii.htm - this photo .
