I'm new to this place. I've been messing around trying to make my pictures transparent because I'm a loser and all I have is MSPaint and Gimp which I barely know how to work.
So here's my question.
Do you absolutely have to have a small image to upload in Pixel Joint? And upon clicking on the small image, will you immediately be directed to the entire image?

Like previews on dA right?

Yes and yes.
Small (previews) are needed if you piece is bigger than 100 pixel in any direction. The preview image then is smaller than 100x100 px, duh!

PJ IS NOTHING LIKE DA!!!

Transparency on the web is only possible through the .GIF format.  Unfortunately, MSPaint does not have the ability to save transparent .GIF images.  I've never used Gimp, but from the screenshots on it's site, it looks advanced enough to have a transparency feature.

And your notions about the previews is correct.  A 'detailed' image is the image that is displayed before someone clicks on it, and it can be at most 100x100 size.  There is a field to give a bigger version of the image if the size goes past 100x100, that is whats shown after someone clicks the smaller image to view the complete image.

FYI, it's alright to have just MSPaint. I've seen a lot of awesome pixellers who just use MSPaint. That's the cool thing about pixel art. It doesn't take a 500 dollar program.

WRONG!

LOL, atleast on IE that is true.  PNG is transparent too, but Microsoft in all thier infinite wisdom decided to be different frome very other web browser and not have png transparency...  which really sucks because with PNG you can have partial transparency on several colors as opposed to GIF where you can only have one color trans...  So with png you can have shadows and AA.

PNG, thanks to IE, is not suggested for trans, however...  Because since +60% of the general surfing population do not use a standards compliant browser...

Now you know, beam me with tomatoes...

Some pieces are easier to quickly except or decline than others. Some can pend for quite a long time until a decision is made by one or more of the mods. While you're waiting you can always pixel more...!

So you go through them all one at a time?
Wow.
Well then I'll just practice more pixeling.

See unlike dA where you can submit whatever crap you want.
Here we have standards for art quality.

So a mod sifts through each art piece, and denys and accepts as he/she sees fit.
