Hello, what is the best method to obtain an effective AA on sprites moving on a changing background, for example in side-scrolling games? Is the use of semi-transparent colors the only option?

Not possible in a true 100% pixel game. The solution for this called 'selout' and is a way to highlight the lineart and diffuse other areas. Most find selout the most complicated technique to get right but its doable. The color pokeman games for GB use this very effectively.

Yeah, it would be sel-out. Though to be honest, results are a bit ugly aesthetically most of the times.

Sel-out is generally considered bad form, unless the luminescence of your background is rigidly consistent it isn't going to work. Try omitting AA on the sprite's exterior, it shouldn't be necessary really. Otherwise use alpha-transparency.
