Well yeah the title is weird, me and a couple of friends are working on a project, and they need a block of jello but i have no idea how to approach this.

first get get the shape right . something like this maby (very bad!) and start from there

Noooo, thats just the style they wanted.
What im supposed to be making is a jello block as the game is in platform view.

Hmm, maybe im not explaining myself well.
Im just trying to make a tileset (block) for the character to move around on.
And he said the view should just be from the side, at the size of 200x50.

So, what you want is not a gelatinous character, but a gelatinous ground block?
Well, a block of jelly viewed like a platform ground is not very recognizable, I suggest you to make it shimmer a lot, and a semi-trasparent effect can do the trick.
